James Jr. attended Tuskegee University for four years (2016–19), participating in all forty-three games while starting at defensive tackle each of those years. In his junior and senior years, he led the team as captain and received the Golder Tiger Character Award.

In his second season, he recorded 62 career tackles, 19 TFLs, 7.5 sacks, and a touchdown from a fumble recovery.
